Recently purchased a Hornady lock-n-load ap and I am needing help determining what powder to use. I keep hearing trailboss does not meter well through the powder drop with the pistol or rifle rotor. New to this round and press. Anyone with 45 colt experience and this press, help greatly appreciated.

I'm not familiar with the press but I can tell you Trail Boss does not meter well in mine. It's -/+ .1 which is a .3 variance. Probably fine for trail boss though. Trail boss is a good powder to learn on though. Although no powders are "safe" it's hard to make a dangerous load with it. Not that you can't if you try.
If you are shooting one of the Italian jobs like pietta or uberti trail boss is excellent. Or an actual colt SAA.
I switched to unique after not getting consistent pass thrus on full power trail boss loads on pigs.
Although I do not share load info for safety reasons a 250gr RNFP hard cast with a stout load of unique will pass thru most anything at close range. Including an 8"x8" oak post out of my lever at 25 yards.
I know this not the info you ask for but just sharing my experiences with the 45 colt.

I am loading 45 with a Hornady LNL too. I believe that I am using Power Pistol powder for them and for 9mm. You really need to focus on ball powders with a progressive press in my opinion.

No experience with the press, but I load my 45 colt plinking ammo with HS-6. Light charges make a pound go a long ways.

Titegroup is another good one for light loads. It and H110 are about all you'll ever need.

I use the older version of the LnL which is the Pro-jector. It works the same, just doesn't have some of the refinements of the LnL, such as the bushings. You did not mention whether you are loading jacketed bullets or cast, so here are my recommendations for both.
For cast bullets: Titegroup, W 231/HP-38, A-5
For jacketed: W296/H110, HS-6, A-5
These all meter with good consistency on my machine.
